To enable, disable, or change the sound played when a new email arrives, type /"Sounds/" into the Start menu and select the first option (Change system sounds). In the window that appears, click on the Sounds tab. then scroll down to New Mail Notification and click on it.

When Mac Mail is set to automatically download new messages (this is the default setting, but you can change how often Apple Mail will automatically check for emails), this will give you a visual and audible signal that new Emails are ready to send. read. The Mail icon in the dock will display the number of unread messages overlaid and the new email sound will play (whether your speaker volume is muted or not, you just won't hear it in this case).
